一、無(wú)色片
染色結(jié)束后，切片中見不到任何陽(yáng)性信號(hào)。這是常規(guī)工作中比較常見的現(xiàn)象，出現(xiàn)這種現(xiàn)象，有兩種可能：1、真陰性結(jié)果：整個(gè)染色過(guò)程沒有出現(xiàn)問題，組織或細(xì)胞確實(shí)不表達(dá)與抗體相關(guān)的抗原。2、假陰性結(jié)果：即此陰性結(jié)果不是真實(shí)的反映。假陰性結(jié)果又可分為兩種情況：（1）切片中根本就不包含所預(yù)期檢查的組織或細(xì)胞。出現(xiàn)這種情況，要麼是病理醫(yī)生選擇錯(cuò)了切片或抗體選錯(cuò)了，要麼是技術(shù)員選錯(cuò)了蠟塊。獲得正確的切片進(jìn)行染色是獲得正確結(jié)果的前提。由此表明：制作出合格的免疫組化切片不僅僅是技術(shù)員的事，病理醫(yī)生也起著*的作用。（2）染色過(guò)程中的某一或某些環(huán)節(jié)出了問題。比如，組織未進(jìn)行抗原修復(fù)，有的組織必須經(jīng)過(guò)抗原修復(fù)才能檢測(cè)抗原表達(dá)；或選用了只能用于冰凍組織而不能用于石蠟包埋組織的抗體；或一抗失效，雖然抗體失效在理論上是一個(gè)逐漸的過(guò)，但偶爾也遇到突然失效的情況，抗體長(zhǎng)期不用和/或已超過(guò)有效期是主要的原因。也可見于染色過(guò)程中漏掉了某一環(huán)節(jié)，如忘記加二抗或三抗，或用了兩次二抗而缺少了三抗，或配制DAB時(shí)少了過(guò)氧化氫。為了避免這種簡(jiǎn)單的錯(cuò)誤，有一種簡(jiǎn)單的方法：在三抗孵育結(jié)束時(shí)，將切片上的三抗甩在一張白紙上，在將配制好的DAB 滴一滴在白紙的三抗上，觀察是否出現(xiàn)棕色。如果出現(xiàn)了，證明三抗和DAB的配制過(guò)程沒有錯(cuò)誤。如果這種DAB再滴到切片上沒有出現(xiàn)任何陽(yáng)性信號(hào)，問題一定是出在三抗以前。如果紙上不出現(xiàn)棕色反應(yīng)，問題肯定在三抗或DAB的配制過(guò)程。這種簡(jiǎn)單方法能迅速的幫助我們查找出現(xiàn)問題可能的原因。
